Στο κέντρο της σύνθεσης, εικονίζεται καθιστός σε περίτεχνα διακοσμημένο θρόνο ο Άγιος Νικόλαος, προστάτης της Μονής, ο οποίος φορά πλήρη αρχιερατική στολή, ευλογεί με το δεξί χέρι και κρατά κλειστό Ευαγγέλιο στο αριστερό. Επάνω, σε νεφέλες, ο Χριστός και η Παναγία του προσφέρουν τα σύμβολα του αξιώματός του, το Ευαγγέλιο και το ωμοφόριον. Γύρω του, σε δύο στήλες, απεικονίζονται έξι σκηνές με θαύματα του Αγίου. Στην κάτω ζώνη εικονίζεται η παραθαλάσσια χτισμένη σε απότομα βράχια τειχισμένη Μονή Γρηγορίου με το Καθολικό της, το καμπαναριό και τη φιάλη της. Διάφορα βοηθητικά κτήρια συμπληρώνουν το τοπίο, όπως κάτω δεξιά το κοιμητήριο και αριστερά ο αρσανάς. Δύο πλοία καταπλέουν στη Μονή, ενώ διακρίνεται στη θάλασσα και βάρκα με δύο μοναχούς.

In the centre, the patron of the monastery, Saint Nicholas, sits on a decorated throne, in his Episcopal dress. With his right hand, he is making a sign of blessing, and in his left hand, he is holding a closed New Testament. Above, in the clouds, Christ and the Virgin Mary hand him the relics suited to his rank: the New Testament and the omophorion. Around the central composition, scenes of the six miracles of Saint Nicholas are depicted. On the lower part, a view of the fortified Gregoriou Monastery is represented. The Monastery is depicted in rocky scenery by the sea, with its Katholikon (main church), bell tower, and phiale (building around a fountain). The view is further detailed with the depiction of subsidiary buildings. On lower right, there is a cemetery, and on lower left, there is an arsanas (harbour).
